A degron-mimicking molecular glue drives CRBN homo-dimerization and degradation

This study identified MRT-31619, a degron-mimicking molecular glue degrader that drives cereblon (CRBN) to self-degrade by bridging two CRBN proteins into a homo-dimer that is then ubiquitinated and degraded, with cryo-EM showing two glue molecules forming a helix-like interface between the CRBN copies. Flow Induced Dispersion Analysis (FIDA), run on a Fida 1 instrument alongside cryo-EM, SEC-MALS, NanoBRET and cellular assays, was used to measure formation of the CRBN:glue:CRBN ternary complex in solution and to rank analogs by EC50, confirming that MRT-31619 induces CRBN homo-dimerization while inactive analogs do not. It is an example of using FIDA to detect and rank molecular-glue ternary-complex formation in solution by the size increase upon complexation.
