This study identified GMAP-1, a secreted lipid transfer protein, as essential for maintaining the impermeability of the nematode cuticle in Caenorhabditis elegans, shuttling lipids to seal the cuticular barrier. To confirm GMAP-1 directly binds lipid, the authors used Flow Induced Dispersion Analysis (FIDA) to measure the interaction of fluorescently labeled phosphatidylcholine (NBD-PC) with the purified protein, determining a dissociation constant (Kd) of ~3 µM and resolving the size of the free lipid versus the lipid–protein complex in solution. It is an example of using FIDA to measure protein–lipid binding affinity in solution.
