Advanced tools for characterizing diverse bispecific antibodies formed through fusion of single-domain antibodies onto IgG scaffolds
Andreas Visbech Madsen & Steffen Goletz
This poster demonstrates how researchers at the Technical University of Denmark (DTU) used Flow Induced Dispersion Analysis (FIDA) to evaluate the in-solution binding functionality of bispecific antibodies formed by fusing single-domain antibodies onto IgG scaffolds. By measuring the apparent hydrodynamic radius of these constructs as they bound two different antigens, PD-L1 and HER2, the team confirmed both antigen-binding sites remained functional, with heavy-chain fusion outperforming light-chain fusion. It is an example of using FIDA to assess multi-target binding functionality of engineered antibody formats directly in solution.
