This Fidabio poster presents an automated method for assessing the conformational stability of therapeutic proteins using only nanogram amounts of sample. Flow Induced Dispersion Analysis (FIDA), on a Fida 1 instrument, was used to measure the hydrodynamic radius and intrinsic fluorescence of adalimumab, human serum albumin and rituximab during in-line chemical denaturation with guanidinium chloride, deriving denaturation midpoints (Cm) and unfolding free energies (ΔG°) from a Fidabio unfolding model — all from ~40 ng of protein per data point. It is an example of using FIDA to characterise protein conformational stability and unfolding in solution.
