Building Confidence Early in Drug Discovery: Integrating FIDA and Biacore SPR for Optimal Results
This webinar presents a drug discovery workflow in which Flow Induced Dispersion Analysis (FIDA) and Biacore surface plasmon resonance (SPR) are applied together, presented by Dr. Philip Addis, head of biophysics at Sygnature Discovery, and Dr. Thomas Bedwell of Fidabio. FIDA characterises protein reagents and compounds before interaction analysis, measuring hydrodynamic radius, aggregation, polydispersity, purity and quantification from a fraction of a microlitre of sample in the buffer the assay will use. Biacore SPR then determines binding kinetics and affinity, and a subsequent FIDA measurement characterises the resulting complex, reporting conformational change, oligomerisation or aggregation. The case studies cover membrane protein batch selection by hydrodynamic radius, identification of aggregation in a kinase preparation prior to interaction analysis, a heat-treated serum albumin preparation, and a compound with unexpected solubility behaviour. At Sygnature Discovery both techniques operate alongside affinity selection mass spectrometry within a hit identification pipeline. It is an example of using FIDA together with surface plasmon resonance so that binding data is generated on characterised material.
