Characterization of Molecular Glues - an automated in-solution platform

This Fidabio poster demonstrates how Flow Induced Dispersion Analysis (FIDA) was used to characterise ternary complex formation between the target protein GSPT1, a molecular glue and the E3 ligase adapter CRBN-DDB1. FIDA quantified complex size (hydrodynamic radius), binding affinity and cooperativity directly in solution, and used an orthogonal intensity readout to confirm binding — distinguishing an active molecular glue from lenalidomide, which produced no ternary complex. It is an example of using FIDA to characterise targeted protein degradation and molecular-glue-induced protein–protein interactions.
