This Fidabio poster demonstrates how Flow Induced Dispersion Analysis (FIDA) was used to rank VHH (nanobody)-producing E. coli clones directly from unpurified cell supernatant, measuring both antigen-binding affinity (Kd) and titre in a single assay. By tracking the hydrodynamic radius of a fixed amount of labelled antigen as it binds VHH in the crude supernatant, reverse-titration curves gave the clone concentration and binding affinity together, without any purification step. It is an example of using FIDA to accelerate early clone screening by combining functional affinity data with expression titre in one unpurified-sample assay.
