Researchers developed Fibril Paint peptides that bind amyloid fibrils and recruit the E3 ubiquitin ligase CHIP, tagging the fibrils with ubiquitin to direct them toward degradation by the ubiquitin–proteasome system. Flow Induced Dispersion Analysis (FIDA), on a Fida 1 instrument, was used to measure the binding affinity of the Fibril Paint peptide FP1 for CHIP (Kd of 1.5 µM) and to detect ubiquitination of Tau amyloid fibrils in solution by tracking the hydrodynamic radius, which increased from 1.8 nm to 42 nm upon ubiquitin conjugation. It is an example of using FIDA to quantify protein–protein binding affinity and monitor real-time particle-size changes during enzymatic modification of amyloid aggregates.
