This study introduces FibrilPaint1, a fluorescent peptide that binds Tau amyloid fibrils, and pairs it with Flow Induced Dispersion Analysis (FIDA) to determine Tau fibril length directly in solution. On a Fida 1 instrument, FIDA measured the hydrodynamic radius (Rh) of the FibrilPaint–fibril complexes, turning the readout into a "molecular ruler" that sizes fibrils across a wide range, tracks their growth during aggregation, and detects them at picomolar concentrations in human serum and in patient-derived Alzheimer's, frontotemporal-dementia and corticobasal-degeneration samples. It is an example of using FIDA to size amyloid fibrils and their complexes in biofluids.
