Fully automated characterization of ternary complex formation using flow induced dispersion analysis

This Fidabio poster demonstrates how targeted protein degradation can be fully characterised in a single automated workflow. Flow Induced Dispersion Analysis (FIDA), on a Fida 1 instrument, was used to measure the hydrodynamic radius of a labelled protein of interest as a bifunctional degrader was titrated against the protein and an E3 ligase, revealing binary and ternary complex formation as a bell-shaped binding curve. From these size measurements FIDA derived dissociation constants for both the protein–degrader and degrader–E3 interactions plus a cooperativity factor of 14.7, using only nanoliters of sample. It is an example of using FIDA to quantify multi-component complex formation and binding cooperativity in solution.
