High Throughput detergent screening for membrane proteins using nanoliters of sample

This poster, led by the National Physical Laboratory (NPL) with Fida Biosystems, demonstrates how Flow Induced Dispersion Analysis (FIDA), on a Fida 1 instrument, was used to screen 11 detergents for their ability to solubilise a GFP-tagged membrane protein (GFP-LacY) directly in crude cell lysate, measuring solubilisation efficiency and polydispersity (PDI) from just 1.5 µL of sample. The FIDA-based detergent ranking matched fluorescence size-exclusion chromatography (FSEC) while cutting screening from ~20 hours to under 3 and sample volume nearly 1,000-fold. It is an example of using FIDA to rapidly screen and characterise membrane protein samples ahead of structural biology work.
