This study identified how α-ketoglutaric acid (AKG) delays senescence in mesenchymal stem cells, pinpointing the ribosomal protein RPS23 as its direct target and validating AKG as a small-molecule agonist with anti-aging potential. Flow Induced Dispersion Analysis (FIDA), on a Fida Neo instrument, was used to measure the binding affinity (Kd) of AKG to the recombinant target protein by titrating AKG against it and tracking changes in the protein's hydrodynamic radius and intrinsic fluorescence, fitting a 1:1 interaction model. It is an example of using FIDA to quantify small-molecule–protein binding affinity in solution under native conditions.
