Pharmacological modulation of septins restores calcium homeostasis and is neuroprotective in models of Alzheimer’s disease

Published in Science, this study identifies septins — cytoskeletal proteins — as a drug target in Alzheimer's disease and characterises a small molecule, REM127, that restores neuronal calcium balance and is neuroprotective in disease models. Flow Induced Dispersion Analysis (FIDA) was used to confirm and quantify how tightly REM127 binds its target: measuring binding of the compound to purified human septin isoforms (SEPT6, SEPT7 and SEPT2), label-free and without immobilisation. FIDA detected selective, high-affinity binding to SEPT6 in the low-nanomolar range, with no measurable binding to SEPT7 or SEPT2 — corroborating the compound's proposed mechanism of action.
