This Fidabio poster demonstrates how Flow Induced Dispersion Analysis (FIDA), on a Fida 1 instrument, was used to rapidly characterise multiple parameters of liquid–liquid phase separation (LLPS): dilute-phase concentration, droplet-formation kinetics, amyloid fibril maturation and modulator binding affinity. Using only microliters of sample, FIDA tracked the aberrant liquid-to-solid transition of α-synuclein into amyloid fibrils and showed how single-stranded DNA disrupts Ddx4n1 droplet formation by binding with a dissociation constant of ~51 µM. It is an example of using FIDA to characterise biomolecular condensates and phase-separating protein systems.
